Holder David Nalbandian has gone out of the Legg Mason Tennis Classic, beaten in straight sets by American James Blake.
Former Wimbledon finalist Nalbandian, seeded eighth for the Washington tournament, crashed out 6-2 6-4 to 31-year-old Blake in just 73 minutes on Tuesday.
Blake, who won his first ATP Tour title at the same event in 2002, allowed his Argentine opponent just three break points, all of which were saved.
Next up for the former world number four is compatriot John Isner, after the 26-year-old eased past Tobias Kamke of Germany 6-1 6-3.
Isner, ranked 35th in the world, scored four breaks of serve in the match and wrapped up victory in just 66 minutes.
Meanwhile, Jarkko Niemenen notched up his 300th win on the ATP Tour with a 7-5 6-4 success against Russia's Igor Kunitsyn.
The 30-year-old Finn turned pro in 2000 and has won one title at the sport's highest level -- the 2006 Heineken Open in Auckland.
The former world number 13 will face either the Czech Republic's Radek Stepanek or American Wayne Odesnik in the third round.
Home crowd favorite Ryan Harrison edged a hard-fought match with Germany's Mischa Zverev 6-4 1-6 6-1.
Harrison, 19, will face third seed Viktor Troicki, a finalist at the William H.G. FitzGerald Tennis Center in 2008, in the next round.
Elsewhere, the top three seeds all survived their opening matches at the WTA event in Carlsbad, California.
Two-time major finalist Vera Zvonareva advanced courtesy of a 6-4 7-5 victory over veteran American Jill Craybas.
Russian Zvonareva, ranked third in the world, secured one break of serve in each set against her 37-year-old opponent to progress.
Second seed Andrea Petkovic is also safely into the third round after a 6-3 6-4 defeat of qualifier Alexa Glatch.
Petkovic, currently ranked 11, is guaranteed to become the first German woman since Anke Huber in 2000 to hold a position in the world's top 10 next week.
Agnieszka Radwanska, a beaten finalist in 2010, battled hard to eliminate Britain's Elena Baltacha.
Third seed Radwanska saved four set points in the second set to eventually emerge with a 6-0 7-6 triumph.
